Sequencing and genotyping

The Lincoln University Sequencing Facility, located within the Bio-Protection Research Centre operates an Applied Biosystems 3130xl Genetic Analyzer, with a 50cm array and POP7 installed as the standard platform.

We offer Full Sequencing, Capillary Separation and Genotyping services five days a week. Results are available 24-48 hours after receipt of samples.

Full sequencing
Full sequencing consists of fluorescent labelling of products (using cycle sequencing PCR) and clean-up of products to remove unincorporated fluorescent ddNTPs, capillary separation and analysis of sequencing results. It is the client's responsibility to prepare, purify, and quantify DNA products, before submitting products for sequencing.

Capillary separation
Capillary separation consists of clean-up of products to remove unincorporated fluorescent ddNTPs, capillary separation and analysis of sequencing results. It is the client's responsibility to prepare, purify, quantify and fluorescently label DNA products, before submitting products for sequencing.

Genotyping
Genotyping (Microsatellite/Fragment Analysis) involves incorporation of a size standard with the fluorescently-labelled DNA product prior to capillary separation. It is the client's responsibility to amplify their template using one or more fluorescently labelled primer.

The default calibration for genotyping uses the DS-33 Matrix Standard calibrated for Dye Set G5 (6FAMTM, VIC®, NEDTM, PET®, LIZ®) Calibration for alternative Dye Sets can be arranged if required. Size Standards held in stock are: LIZ-500, LIZ-600 and LIZ-1200. Additional size standards can be ordered as required.
